B-Methylated-II delivers a balanced combination of two bioactive B vitamins-L-Methylfolate (5-MTHF) and Vitamin B12 as methylcobalamin- formulated to support core methylation pathways, neurological function and homocysteine metabolism.

Unlike high-dose, single-nutrient formulas, this product is designed for daily foundational support, providing active forms that are immediately usable by the body without reliance on enzymatic conversion.

Bioactive Folate & Active B12 for Brain and Nervous System Support

L-Methylfolate (5-MTHF) is the biologically active form of folate used directly in the body, including within the central nervous system. Vitamin B12 (as methylcobalamin) works synergistically with folate in one-carbon metabolism, a pathway essential for neurotransmitter synthesis, nerve function and cognitive processes.

At 3 mg of L-Methylfolate paired with 3,750 mcg of methylcobalamin, B-Methylated-II provides meaningful nutritional support for brain chemistry and nervous system function- without the intensity associated with therapeutic-strength dosing.

Supports Healthy Homocysteine Metabolism & Cardiovascular Function

Folate and vitamin B12 are key cofactors in the remethylation of homocysteine to methionine. Maintaining healthy homocysteine levels is important for cardiovascular, neurological and cellular health.

By supplying both nutrients in their active, methylated forms, B-Methylated-II supports efficient homocysteine metabolism and overall methylation balance as part of a daily wellness routine.

Designed for Individuals Who Prefer Active B Vitamins

Some individuals choose methylated forms of B vitamins due to personal preference or reduced efficiency in converting conventional forms such as folic acid or cyanocobalamin.

B-Methylated-II provides:

  • L-5-methyltetrahydrofolate (5-MTHF) instead of folic acid
  • Methylcobalamin instead of synthetic B12 forms

This approach ensures the nutrients are ready to use upon absorption, supporting consistent metabolic activity without additional conversion steps.

Balanced, Everyday Methylation Support

The 3 mg L-Methylfolate / 3.75 mg methylcobalamin ratio is intentionally designed to provide supportive-not excessive-methylation activity. This makes B-Methylated-II suitable for:

  • Daily foundational methylation support
  • Long-term nutritional use
  • Individuals who may not require high-dose, practitioner-level protocols

Because methylation needs vary, some individuals prefer starting with a balanced formula before considering higher-dose, single-nutrient options.

Clinically Relevant, Complementary Benefits

How much should you take?

Methylation needs vary widely between individuals. For this reason, many practitioners recommend a “start low, go slow” approach when introducing methylated B vitamins, especially for those new to methylation support or sensitive to methyl donors.

Suggested ramp-up approach:

  • Week 1: 1 chewable tablet every other day
  • Week 2: 1 chewable tablet daily
  • Week 3+: Continue 1 chewable tablet daily for ongoing nutritional support

This gradual approach allows your methylation pathways and neurotransmitter systems to adapt smoothly to increased availability of active folate and vitamin B12.

Why the ramp?

L-Methylfolate and methylcobalamin participate directly in methyl group transfer and neurotransmitter synthesis. In some individuals, introducing active forms too quickly may temporarily affect energy, focus or mood as metabolic pathways adjust.

A phased schedule supports:

  • Smoother adaptation of methylation activity
  • Balanced neurotransmitter support
  • Improved long-term tolerance

Clinical and biochemical research supports gradual titration of methylated B vitamins to promote comfort and adherence.
(PubMed: PMC3658370)

Timing and administration

  • Take after meals
  • Can be chewed, swallowed or allowed to dissolve in the mouth
  • Some individuals prefer morning use due to B12’s role in energy metabolism

Consistency is more important than timing-choose a routine you can maintain.

Consistency matters:

Because methylation influences multiple downstream pathways, benefits may build gradually over time.

Many users report:

  • Subtle improvements in mental clarity or energy within 1-3 weeks
  • More noticeable benefits with consistent use over 4-8 weeks

Long-term nutritional support is associated with more stable methylation balance.
(PubMed: PMC12286010).

If you’re sensitive

If you are sensitive to methylated nutrients or new to methylation support:

  • Maintain every-other-day use for 2-3 weeks before increasing
  • Temporary sensations such as mild restlessness or changes in focus may occur as pathways adjust

If symptoms persist or become uncomfortable, discontinue use and consult your healthcare professional.

Using with other supplements or medications

  • If using additional B-complex vitamins, monitor total intake to avoid redundancy
  • If taking prescription medications or managing a health condition, consult your clinician before use

While methylated B vitamins are nutritional supplements, they can influence biochemical pathways relevant to medication metabolism.
(PubMed: PMC12276071)

Why Methylated B12 + L-Methylfolate Matters

Methylation is a foundational biochemical process that influences DNA synthesis, gene expression, neurotransmitter production, red blood cell formation and homocysteine metabolism. At the center of this system are folate and vitamin B12, which function together in one-carbon metabolism to transfer methyl groups required for normal cellular activity.

When folate or vitamin B12 status is suboptimal, methylation efficiency can decline. This may affect neurological function, cardiovascular health, energy metabolism and cellular repair processes. Because conventional forms such as folic acid and cyanocobalamin require multiple enzymatic conversion steps, some individuals prefer bioactive forms that are ready for immediate use.

L-Methylfolate (5-MTHF) is the biologically active form of folate used directly by cells, including within the central nervous system. Methylcobalamin is an active form of vitamin B12 that supports methyl group transfer and nervous system integrity. Together, they provide direct nutritional support for methylation pathways without reliance on conversion efficiency. (PMC3888748)

Important caveat: Methylated B vitamins support normal physiological function but are not intended to treat disease. Individuals with medical conditions or those taking prescription medications should consult a healthcare professional before use. Research on methylation nutrients is extensive, though outcomes vary based on individual genetics, diet and overall nutrient status.

Balanced Methylation Support for Brain Function, Cellular Energy & Everyday Resilience.

Unlock Clinically-Informed Cellular Protection

Formulated as a once-daily chewable tablet, B-Methylated-II delivers foundational support for one of the body’s most essential biochemical systems: methylation. This process underpins DNA synthesis, neurotransmitter production, red blood cell formation and homocysteine metabolism-functions that influence how you think, feel and generate energy each day.

When methylation pathways are under-supported, you may experience mental fog, low energy, poor stress tolerance or reduced cognitive clarity. Over time, inefficient methylation can affect cellular balance and overall metabolic efficiency.

Supports Efficient Methylation & Cellular Function

Methylation is a core biological process that influences DNA synthesis, gene expression, detox pathways and cellular repair. When methylation is functioning efficiently, cells are better able to regulate normal turnover, maintain genomic stability and support long-term metabolic balance.

By providing active folate and active vitamin B12 together, B-Methylated-II supports the body’s ability to sustain methyl group availability needed for these foundational cellular processes. Consistent daily support helps maintain steady methylation activity rather than sharp spikes or deficiencies that can disrupt downstream systems.

Evidence-based insight:
Clinical and biochemical research confirms that folate and B12 are essential cofactors in one-carbon metabolism and DNA methylation, supporting normal cellular function and repair.
(PMC3262611)

Supports Cellular Energy & Red Blood Cell Formation

Energy production depends on efficient cellular metabolism and oxygen delivery. Vitamin B12 and folate are required for red blood cell formation and DNA synthesis in rapidly dividing tissues.

When these processes are supported:

  • Oxygen transport remains efficient
  • Cellular energy metabolism is maintained
  • Fatigue related to nutrient insufficiency is less likely

B-Methylated-II provides both nutrients in forms the body can use immediately, supporting normal energy metabolism as part of daily nutritional intake.

Evidence-based insight:Human studies show that adequate folate and vitamin B12 intake supports red blood cell production and energy metabolism, particularly in tissues with high turnover (PMC3257642).

Supports Brain, Cognitive & Nervous System Health

The brain is highly sensitive to disruptions in folate and B12 availability. Active folate is required to maintain brain folate levels involved in neurotransmitter synthesis, while vitamin B12 supports nerve integrity and myelin maintenance.

Adequate availability of both nutrients supports:

  • Normal neurotransmitter balance
  • Cognitive processes such as focus and mental clarity
  • Nervous system signaling and resilience

By supplying methylated forms that are directly usable, B-Methylated-II supports neurological function without relying on conversion from synthetic vitamin forms.

Evidence-based insight:L-Methylfolate is actively transported across the blood-brain barrier and, together with vitamin B12, supports neurotransmitter pathways and cognitive health. (PMC6750952).

Supports Healthy Homocysteine Metabolism & Cardiovascular Balance

Homocysteine is an intermediate amino acid that must be efficiently recycled for cardiovascular and neurological health. Folate and vitamin B12 are essential for converting homocysteine back into methionine, a key step in methylation and metabolic balance.

Supporting this pathway helps maintain:

  • Normal homocysteine metabolism
  • Vascular and endothelial function
  • Long-term cellular and neurological health

Providing both nutrients together supports this interconnected system more effectively than isolated supplementation.

Evidence-based insight:Clinical research demonstrates that bioactive folate and vitamin B12 contribute to maintaining homocysteine levels within a healthy range (PMC10494275).

Clinically Studied, Backed by Real Science

Methylated folate (5-MTHF) and active vitamin B12 (methylcobalamin) are among the most researched nutrients involved in one-carbon metabolism, a foundational biochemical system that regulates methylation, DNA synthesis and cellular signaling. Human and mechanistic studies consistently show that adequate availability of these nutrients is essential for maintaining normal metabolic balance, neurological function and long-term cellular health.

Supports Methylation & DNA Regulation

Foundational support for gene expression, cellular repair & metabolic balance

Methylation is a core biochemical process that helps regulate gene expression, DNA synthesis and cellular repair. Folate and vitamin B12 are essential cofactors in the methionine cycle, where they support the conversion of homocysteine into methionine and the production of S-adenosylmethionine (SAM)-the body’s primary methyl donor.

Adequate methyl group availability is required for:

  • DNA and RNA synthesis

Understanding DFE Labeling for Methylfolate

What the New Numbers on Your Label Mean - and Why They Haven’t Changed Your Dose

When it comes to folate supplements, labels can be misleading - not because the formulas are different, but because the rules have changed. In this video, Jamie from Methyl-Life® explains the FDA’s new requirement to use Dietary Folate Equivalents (DFE) and what that means for how folate is reported on supplement labels.

What’s this video about?: How the new FDA labeling standards affect folate supplements - and why your formula hasn’t changed, even if the number on the label looks much higher.

B-Methylated II: Pure B12 & Bioactive B9 for MTHFR & Neuropathy | Methyl-Life® Explained

The benefits of using already-converted, bioactive B9 and B12—ready for your cells to use immediately

Unlock better methylation support with B-Methylated II from Methyl-Life®, introduced by founder Jamie Hope. In this video, Jamie explains how B-Methylated II is specially formulated for people with MTHFR and methylation challenges, delivering the most biologically active forms of B9 (methylfolate) and B12 (methylcobalamin) for maximum effectiveness.

B-Methylated II offers a cost-effective, over-the-counter alternative to prescription products like Metanx, but without the risk of high-dose B6 toxicity

Why Folate Labels Now Use DFE

The FDA now requires that all folate-containing supplements list folate content in mcg DFE to account for how much folate is actually absorbed by the body, based on food-based folate sources.

  • Folic acid × 1.7 = mcg DFE
  • Methylfolate × 1.7 = mcg DFE

This change reflects differences in bioavailability. Synthetic folic acid, for example, has ~85% bioavailability when taken with food. Natural, whole-food-based folate has ~50% bioavailability. The DFE unit helps consumers understand how much usable folate they’re getting compared to whole foods.

Example:

  • A supplement with 1,000 mcg folic acid will now be labeled as 1,700 mcg DFE.
  • A supplement with 15,000 mcg L-Methylfolate will now show 25,500 mcg DFE (or more, depending on the purity and form used).

But this is not an increase in the dose - just new math to reflect body-usable folate content.

Why This Matters for Methyl-Life® Users

Methyl-Life® uses L-5-Methyltetrahydrofolate (L-Methylfolate) - a bioactive form of folate that does not require MTHFR activity. Even though it’s more effective and better absorbed than folic acid, the FDA still requires methylfolate products to use the same 1.7 multiplier for DFE calculations.

So if your bottle used to say 15,000 mcg of L-Methylfolate and now it says 25,500 mcg DFE, the dose hasn’t changed - the math has. You're still receiving 15 mg of pure L-Methylfolate, now labeled using the updated DFE mcg format.

Why the DFE Conversion Matters Less Than the Form

DFE was designed around folic acid absorption. But Methyl-Life® uses methylfolate, which already comes in its active form and offers superior uptake - especially for those with MTHFR gene mutations.

This is supported by evidence showing that L-Methylfolate:

So while labeling has changed, nothing has changed in your supplement’s clinical performance.

Recap: DFE Labeling Doesn’t Change the Dose

  • DFE stands for Dietary Folate Equivalent, a unit created to reflect folate’s usable potency compared to whole-foods-based absorption levels
  • The formula is exactly the same - only the label has changed
  • Methyl-Life® continues to deliver clinically studied L-Methylfolate at the same high potency, now labeled to meet FDA guidelines
  • You do not need to recalculate your dose or change your usage
