A 2024 animal study found that high-dose methylfolate may protect against neurodevelopmental behaviours resulting from exposure to pyrethroid, a type of pesticide. Pyrethroid exposure disrupts folate metabolism in the brain and may contribute to behavioral disorders such as autism. 

Methylfolate is the biologically active form of folate and can bypass disruptions to the breakdown of folate in the body, whether caused by MTHFR or by DPE exposure. In this study, high-dose supplementation with methylfolate dramatically reduced the behavioural disorders caused by folate issues, preventing deficits in the social, communication, cognitive, and locomotor areas of the brain that are linked to autism.¹⁹

Why take a methylfolate supplement?

Folate is required for numerous critical functions of the brain and nervous system. It is essential for DNA/RNA synthesis, maintaining healthy homocysteine levels, and for proper fetal development during pregnancy. 

Folic acid is a synthetic form of folate. It can only be used by the body after it has been converted to active folate by an enzyme called methylenetetrahydrofolate reductase (MTHFR)¹³.

Who is affected by MTHFR mutations?

The MTHFR mutation is much more common than many people realize (it’s estimated around 50% of the global population have at least 1 of 2 common variants).  Approximately 25% of the global population are carriers of the MTHFR mutation C677T, with Hispanics making up the highest proportion (47%), East Asians (30%), South Asians (12%) and Africans (9%).  Another 25% of the global population have the MTHFR mutation A1298C, which is highest in South East Asians (42%) and Europeans (31%). The frequency of the 1298A>C polymorphism in Hispanics and Africans is 15%¹⁴

An increasing number of studies have shown links between those with MTHFR polymorphisms and an increased risk for high levels of homocysteine, depression, and reduced antidepressant effectiveness¹⁵.  MTHFR is also linked to elevated homocysteine levels, which can cause serious vascular diseases, dementia, infertility, and other chronic illnesses¹⁶.

Basically, yes. In the chemistry world, L stands for the “Left” isomer, which is the same as the “(6S)” isomer – the terms are synonyms (and this naming convention is used interchangeably by various companies in the industry). In the case of methylfolate, that L or (6S) isomer is the active isomer of 5-MTHF that the body can actually use (conversely, D or (6R) refers to the inactive isomer of 5-MTHF, which isn’t the exact form the human body needs). Not all methylfolate is created equal. Why Folate Labels Now Use DFE

The FDA now requires that all folate-containing supplements list folate content in mcg DFE to account for how much folate is actually absorbed by the body, based on source.

  • Folic acid × 1.7 = mcg DFE
  • Methylfolate × 1.7 = mcg DFE

This change reflects differences in bioavailability. The folic acid nutrient, for example, has ~85% bioavailability when taken with food. However, natural food folate has only ~50% bioavailability. The DFE unit helps consumers understand how much “usable folate” they’re getting as if it were compared to eating folate from whole foods.

Example:

  • A supplement with 1,000 mcg folic acid will now be labeled as 1,700 mcg DFE.
  • A supplement with 15,000 mcg L-Methylfolate will now show 25,500 mcg DFE (or could show more, depending on the salt form used and its potency calculation).

NOTE:  this is not an increase in the dosage amount — just new math to reflect usable folate content from the same formula.

Why This Matters for Methyl-Life® Users

Methyl-Life® uses L-5-Methyltetrahydrofolate (L-Methylfolate) — a bioactive form of folate that does not require MTHFR conversion. Even though it’s more effective and better absorbed than folic acid, the FDA still requires methylfolate products to use the same 1.7x multiplier for DFE.

So if your bottle used to say 15,000 mcg of L-Methylfolate, and now it says 25,000-30,000 mcg DFE, just know that neither the formula nor the dose has changed — only the math has. You're still receiving 15 mg of pure L-Methylfolate, now labeled using the updated DFE format.

Why the DFE Conversion Matters Less Than the Form

DFE was designed around folic acid absorption. But Methyl-Life® uses methylfolate, which already comes in its active form and offers superior uptake — especially for those with MTHFR gene mutations.

"The product itself is fine, but I would like to share my experience that may help others. I have the double MTHFR gene mutation. Everything I read said to take these supplements. When I began taking them, my anxiety went through the roof!!! I was angry and anxious. I read about over methylation and I was experiencing this (even on a lower dose). Sometimes people don’t react well to methyl supplements. So just listen to your body. If you experience these side effects, you may want to consider Folinic acid. I hope Methyl Life will make a Folinic acid supplement."

We're glad you tried our supplements and took the time to write a review. You're absolutely right, not everyone responds well to methylated forms of nutrients, especially when it's the first thing they try. Have you taken any NAC by chance? This is often a good thing to try, it will definitely help benefit those who are overmethylating - and for many taking it along with the methylated forms can really be beneficial (and potentially even allow them to tolerate some of the methyl forms). Folinic acid does not get around the MTHFR mutation, so for that reason Dr. Neil Rawlins does not suggest it as best if you have an MTHFR mutation, however, it can work in conjunction with methylfolate for some (I know Autism Speaks was doing a trial with it). Folinic acid is technically Leucovorin and so can be classified as a drug to treat cancer in some cases, we are still researching how possible it is for us to leverage it in our products legally. And it's worth noting that Dr. Neil Rawlins suggests methylfolate instead of Leucovorin in many cases. Again, thank you for taking the time to leave us feedback.
